You probably hit this issue: https://git.ispconfig.org/ispconfig/ispconfig3/-/issues/6389 To fix that, go to system > server config > mail, there you find the spamfilter setting. switch this setting from amavis to rspamd, click save, switch back to amavis, click save. Wait a minute and test again. This will rewrite the filter setup and fix the issue.
Have you tried logging in to roundcube and sending e-mail to yourself. Does that work? If not, what is in mail log? Is your setup an e-mail server with port 25 closed, and you have set up relaying? How did you do that? Use the e-mail ID number of the failed attampt to find all entries about that in recent /var/log/mail.* files.
