I am looking to have one Name Server Mirror two different master Name Servers. Is this possible, or should I set up a second slave server for the mirroring of the second Name Server?
You can't do that with the ispconfig mirror function as a mirror can have only one master. What you can do is that you use the "dns slave records" in the dns module instead, Florian has written a module for that https://git.ispconfig.org/ispconfig/Modules/tree/master/dns_slave_auto
